A polysaccharide fraction extracted from Pleurotus ostreatus mycelial biomass inhibit Sarcoma 180 tumor Anais da ABC (AABC)
WISBECK,ELISABETH; FACCHINI,JEAN M.; ALVES,ENDI P.; SILVEIRA,MARCIA L.L.; GERN,REGINA M.M.; NINOW,JORGE L.; FURLAN,SANDRA A..
ABSTRACT Fungi of Pleurotus genus have attracted a great interest due to their medicinal properties such as anti-inflammatory, antimicrobial and antitumor. These properties are attributed mainly to polysaccharides synthesized by Pleurotus. This work aimed to study the mycelial growth of P. ostreatus in submerged culture, evaluating the influence of the initial concentration of substrate (20 and 40 g/L of glucose) and the pH (4 and 6) on kinetic parameters of production of biomass. The effectiveness of different doses (10, 30 and 50 mg/kg) of a mycelium polysaccharide fraction extracted from P. ostreatus in reducing Sarcoma 180 development in mice was also verified. In the range of this study, maximum concentration of mycelial biomass (about 12.8 g/L) was...

A Rapid and Efficient Method for Purifying High Quality Total RNA from Peaches (Prunus persica) for Functional Genomics Analyses Biol. Res.
MEISEL,LEE; FONSECA,BEATRIZ; GONZÁLEZ,SUSANA; BAEZA-YATES,RICARDO; CAMBIAZO,VERONICA; CAMPOS,REINALDO; GONZALEZ,MAURICIO; ORELLANA,ARIEL; RETAMALES,JULIO; SILVA,HERMAN.
Prunus persica has been proposed as a genomic model for deciduous trees and the Rosaceae family. Optimized protocols for RNA isolation are necessary to further advance studies in this model species such that functional genomics analyses may be performed. Here we present an optimized protocol to rapidly and efficiently purify high quality total RNA from peach fruits (Prunus persica). Isolating high-quality RNA from fruit tissue is often difficult due to large quantities of polysaccharides and polyphenolic compounds that accumulate in this tissue and co-purify with the RNA. Here we demonstrate that a modified version of the method used to isolate RNA from pine trees and the woody plant Cinnamomun tenuipilum is ideal for isolating high quality RNA from the...

Capillary electrophoresis determination of the binding affinity of bioactive sulfated polysaccharides to proteins: study of the binding properties of fucoidan to antithrombin ArchiMer
Varenne, A; Gareil, P; Colliec-jouault, Sylvia; Daniel, R.
The interaction of proteins with polysaccharides represents a major and challenging topic in glycobiology, since such complexes mediate fundamental biological mechanisms. An affinity capillary electrophoresis method has been developed to evidence the complex formation and to determine the binding properties between an anticoagulant polysaccharide of marine origin, fucoidan, and a potential target protein, antithrombin. This method is a variant of zonal electrophoresis in the mobility shift format. A fixed amount of protein was injected into a capillary filled with a background electrolyte containing the polysaccharide in varying concentrations. The effective mobility data of the protein were processed according to classical linearization treatments to...

Characterization and biological properties of sulfated polysaccharides of Corallina officinalis and Pterocladia capillacea Acta Botanica
Ismail,Mona Mohamed; Amer,Mohamed Saleh.
ABSTRACT Red seaweed possess various sulfated polysaccharides (SPs) that could potentially be exploited as bioactive agents for medical and industrial applications. Crude polysaccharides from the red algae Corallina officinalis (SP1) and Pterocladia capillacea (SP2) were extracted and characterized according to their chemical content and their antioxidant, anti-inflammatory, anticoagulant, antibacterial, antifungal, and antifouling activities. The isolated polysaccharides contained low levels of protein and high levels of carbohydrate and sulfate. The extracted SPs were characterized by Fourier-transform infrared (FTIR) and nuclear magnetic resonance (NMR) spectral data and revealed that SP1 is composed of carrageenan, while SP2 is composed of...

Colleters in Chamaecrista (L.) Moench sect. Chamaecrista and sect. Caliciopsis (Leguminosae-Caesalpinioideae): anatomy and taxonomic implications Acta Botanica
Silva,Marinalva dos Santos; Coutinho,Ítalo Antônio Cotta; Araújo,Maicon Nascimento; Meira,Renata Maria Strozi Alves.
ABSTRACT The genus Chamaecrista contains ca. 330 species organized into six sections, of which section Chamaecrista is the second largest (ca.75 species) distributed among six series, while the section Caliciopsis contains only two species. Colleters have been described in the genus Chamaecrista and they show potential taxonomic and phylogenetic significance. They are associated with lubrication, desiccation prevention and protection from microbial attacks of young developing organs. Although six types of colleters have been described for the genus Chamaecrista, there have been no studies focusing on the diversity of colleters in the sections Chamaecrista and Caliciopsis. Samples from developing leaves and flowers of both sections were obtained from...

Comparative anatomy of calyx and foliar glands of Banisteriopsis C. B. Rob. (Malpighiaceae) Acta Botanica
Araújo,Josiane Silva; Meira,Renata Maria Strozi Alves.
Banisteriopsis is considered one of the largest genera of Malpighiaceae with 58 species, of which 47 occur in Brazil. The typical calyx and leaf glands of Banisteriopsis are considered relevant to the adaptive success of Malpighiaceae. Comparative studies of anatomical and histochemical characteristics may reveal similarities and assist in the interpretation of the functions performed by such glands. The present study aimed to describe the anatomy of the calyx and leaf glands of 38 species ofBanisteriopsis that occur in Brazil, and to analyze these structures histochemically in B. campestris, B. laevifolia and B malifolia, using standard methods. Calyx glands differ from leaf glands by possessing an irregular surface that is covered by a thick cuticle that...

Effects of different extracts of the mushroom Agaricus blazei Murill on the hematologic profile of mice with Ehrlich tumor Arq. Bras. Med. Vet. Zootec.
Verçosa Júnior,D.; Ferraz,V.P.; Duarte,E.R.; Oliveira,N.J. F.; Soto-Blanco,B.; Cassali,G.D.; Melo,M.M..
The aim of this study was to investigate the effect of the mushroom Agaricus blazeii Murril (ABM) extracts on the hematological profile of Swiss mice bearing an Ehrlich solid tumor. Three fractions (total extract, polysaccharides, and supernatant) of ABM extracts obtained by four methods (ultrasonic or water bath, at pH 4 or pH 7) were administered to mice over 21 days. Polysaccharide solutions were analyzed by gas and liquid chromatography that showed both mannose and glucose concentrations. The method of extraction influenced the degree of glucose polymerization and the mannose/glucose relationship. The treatment with ABM supernatant at pH 7 and water bath was associated with reduced concentrations of leukocytes and lymphocytes and altered the percentage...

Enhancement of cellular activity in hyperglycemic mice dermal wounds dressed with chitosan-alginate membranes BJMBR
Breder,J.S.C.; Pires,A.L.R.; Azevedo,F.F.; Apolinário,P.P.; Cantaruti,T.; Jiwani,S.I.; Moraes,Â.M.; Consonni,S.R.; Araújo,E.P.; Adams,G.G.; Saad,M.J.A.; Lima,M.H.M..
The use of specially designed wound dressings could be an important alternative to facilitate the healing process of wounds in the hyperglycemic state. Biocompatible dressings combining chitosan and alginate can speed up wound healing by modulating the inflammatory phase, stimulating fibroblast proliferation, and aiding in remodeling phases. However, this biomaterial has not yet been explored in chronic and acute lesions of diabetic patients. The aim of this study was to evaluate the effect of topical treatment with a chitosan-alginate membrane on acute skin wounds of hyperglycemic mice. Diabetes mellitus was induced by streptozotocin (60 mg · kg-1 · day-1 for 5 days, intraperitoneally) and the cutaneous wound was performed by removing the epidermis using...

Etude de la biosynthèse des polysaccharides bactériens : vers la production recombinante d'hétéropolysaccharide ArchiMer
Lebellenger, Lou.
The marine bacteria Vibrio diabolicus produces an exopolysaccharide (EPS) called HE800. The native form of HE800 has already shown biological efficiency in bone healing. It is studied as glycosaminoglycan-mimetic for applications in cosmetic and human health. To enhance biotechnological applications of polysaccharides, the chemical structures should be properly controlled; moreover, the production yield could also be a bottleneck. The genetic cluster of the HE800 EPS biosynthesis has been identified in 2014. However, the biosynthesis molecular mechanisms and regulations remain sometimes unclear. The main objective of this work is to obtain a recombinant production of the HE800 EPS in the common host Escherichia coli, to improve the knowledge of its...

Etude de la réponse immunitaire chez l'huître Crassostrea gigas : caractérisation et fonctions de protéines de reconnaissance aux LPS et d'effecteurs antimicrobiens ArchiMer
Gonzalez, Marcelo.
Research on the immune response of oysters is motivated by the problems of infectious diseases potentially found in the farms. Over the course of the last few years, the development of genomic approaches has allowed us to make headway on characterising genes involved in the cup oyster's (Crassostrea gigas) innate immunity, particularly thanks to various programmes for analysing transcriptome, of type EST (Express Sequence Tag) and subtractive banks. Within the context of this work, over the course of which we have used the data from genomic research and we have also developed biochemical approaches, we were particularly interested in studying the proteins involved in reconnaissance reactions to LPS as well as to antimicrobial effectors. In an earlier part,...

Extraction of Soluble Polysaccharides from Grape Skins Ciencia e Investigación Agraria
Gil Cortiella,Mariona; Peña-Neira,Álvaro.
Obtaining polysaccharides from grape cell walls is an interesting topic for the wine industry given its implications with regards to wine treatments and potential sensory attributes. In the present work, several conditions (i.e., a chelating agent, pH, titratable acidity, extraction time, number of successive extractions, solid to liquid ratio and alcoholic fermentation) were tested to assess their effects on the yield of soluble polysaccharides obtained from grape skins. Among the four tested solvents (i.e., water, tartaric acid, ammonium oxalate, and ethylenediaminetetraacetic acid), aqueous ammonium oxalate was the most effective for polysaccharide extraction but also produced the highest amount of protein impurities. Increasing the number of successive...

Glycoconjugates and polysaccharides of fungal cell wall and activation of immune system BJM
Pinto,M.R.; Barreto-Bergter,E.; Taborda,C.P..
The fate of organochlorine 14C-dicofol in activated sludge process was investigated. Results showed that the major part of radioactivity remained adsorbed on biological sludge. Consequently, its final disposal deserves special attention. The small amounts of dicofol, biotransformed or not, which remained in the treated effluent could contaminate receiving bodies.

Isolation of high-quality RNA from Platycladus orientalis and other Cupressaceae plants Electron. J. Biotechnol.
Chang,Ermei; Zhao,Yuxin; Wei,Qianchun; Shi,Shengqing; Jiang,Zeping.
Platycladus orientalis has a lifespan of several thousand years in China, making it a good plant in which to study aging at the molecular level, but this requires sufficient quantities of high-quality P. orientalis RNA. However, no appropriate methods have been reported for total RNA isolation from P. orientalis leaves. The TRIzol method did not extract RNA, while cetyltrimethylammonium bromide, sodium dodecyl sulfate-phenol, and plant RNAout kit (Tianz, Inc., China) protocols resulted in low yields of poor quality RNA. Isolating total RNA using the Spectrum™ Plant Total RNA Kit (Sigma-Aldrich, St. Louis, MO, USA) resulted in a high-quality product but a low yield. However, the two-step removal of polyphenols and polysaccharides in the improved plant...

Isolation of total RNA from tissues rich in polyphenols and polysaccharides of mangrove plants Electron. J. Biotechnol.
Rubio-Piña,Jorge A; Zapata-Pérez,Omar.
RNA extraction from mangrove tissues can be difficult due to the presence of polyphenolic and polysaccharide compounds upon cell disruption. Besides, a successful RNA isolation from mangrove tissues sometimes can be strain and species specific. Two different methods were used to extract RNA from tissues (stems, leaves and roots) of black mangrove (Avicennia germinans) and white mangrove (Laguncularia racemosa), collected from the mangrove area at Progreso, Yucatan, Mexico. An optimized and modified total RNA isolation method was developed for these recalcitrant species. The protocol is based on the CTAB method including β-mercaptoethanol and PVPP with sequential Phenol/Chloroform/isoamyl alcohol extractions to remove protein, and polyphenols,...

Pharmacological Modulation of Human Mesenchymal Stem Cell Chondrogenesis by a Chemically Oversulfated Polysaccharide of Marine Origin: Potential Application to Cartilage Regenerative Medicine ArchiMer
Merceron, Christophe; Portron, Sophie; Vignes-colombeix, Caroline; Rederstorff, Emilie; Masson, Martial; Lesoeur, Julie; Sourice, Sophie; Sinquin, Corinne; Colliec-jouault, Sylvia; Weiss, Pierre; Vinatier, Claire; Guicheux, Jerome.
Mesenchymal stem cells (MSCs) are considered as an attractive source of cells for cartilage engineering due to their availability and capacity for expansion and multipotency. Differentiation of MSC into chondrocytes is crucial to successful cartilage regeneration and can be induced by various biological agents, including polysaccharides that participate in many biological processes through interactions with growth factors. Here, we hypothesize that growth factor-induced differentiation of MSC can be increased by chemically oversulfated marine polysaccharides. To test our hypothesis, human adipose tissue-derived MSCs (hATSCs) were cultured in pellets with transforming growth factor (TGF)-beta 1-supplemented chondrogenic medium containing either the...
